How to identify the location?

when i used to check any particular text is appearing or not?

When i record it is taking as
Command: Assert Text
Target: //div[2] or //div[3]
Value: The amount of the transaction must be greater than 0

When I manually write Test Case how should i identify this locations (Target).,...

Please suggest me how to do this when i writing manually code?

You can use selenium.IsElementPresent command...
